摘要
目的:通过对湖北汉族2型糖尿病患者进行HLA-DRB1基因分型,探讨此人群与HLA-DRB1的相关性。方法:用序列特异性寡核苷酸探针聚合酶链反应(PCR-SSOP)技术对162例2型糖尿病患者及1 358例健康人群进行HLA-DRB1基因分型。基因频率与Hardy-Weinberg平衡卡方检测使用arlequin软件进行,组间比较用Fisher精确概率法计算P值,检测水准为α=0.05,相对危险系数用RR表示,RR=A(A+B)/C(C+D)。结果:T2DM患者中检出HLA-DRB1等位基因共13种与对照组一致。其中DRB1*08与DRB1*09基因频率显著高于对照组(P均<0.005),有统计学意义,且RR>1,说明DRB1*08与DRB1*09可能是的T2DM易感基因;DRB1*14基因频率显著低于对照组(P<0.005),有统计学意义,且RR<1,说明DRB1*14可能是的T2DM的保护基因。结论:2型糖尿病与HLA-DRB1有一定的关联,但有其自身的特点。
Objective: To explore the association of HLA-DRB1 alleles with type 2 diabetes mellitus of Han population in Hu- bei province by testing HLA-DRB1 genotypes. Methods:In 162 patients with T2DM and 1 358 healthy population,PCR-SSOP methods were applied to determine HLA-DRB1 genotypes. Gene frequency and Hardy-Weinberg balance test was completed by using Arlequin software. Comparison among groups was made using precision probabilistic method ( P, α = 0.05 ) and the relative danger coefficient was expressed by RR. Results:Thirty kinds of HLA-DRB1 alleles were found in the T2DM patients, that was consistent with the control specimens. The gene frequencies of HLA-DRB1 * 08 and DRB1 * 09 were significantly higher than the control specimens, and it is per- haps that the HLA-DRB1 * 08 and DRB1 * 09 are the susceptibility genes of T2DM(P 〈0.005 ,RR 〉 1 ) ; the gene frequency of HLA- DRB1 * 14 was significantly lower than the control specimens, and it is perhaps that the HLA-DRB1 * 14 was the protection gene of T2DM ( P 〈 0. 005, RR 〈 1 ). Conclusion: There was association with HLA-DRBI alleles and T2DM, but that has itself characteristic.
