摘要
探讨MMP-2蛋白及mRNA在胶质瘤细胞中的表达及其在胶质瘤的发生及恶性进展中的作用。采用免疫组化法及原位杂交法检测5例正常脑组织及40例不同级别胶质瘤中MMP-2mRNA蛋白及mRNA的表达情况。正常脑组织中MMP-2mRNA蛋白及mRNA的表达均为阴性。在脑胶质瘤中MMP-2蛋白主要表达于瘤细胞及血管基底膜的内皮细胞的胞浆,其表达是在胞浆中出现棕色或棕黄色染色。在Ⅰ、Ⅱ级胶质瘤标本23例中阳性4例、弱阳6例、阴性13例,阳性表达率为43.48%(10/23),LI值为(5.91±4.78)%,在Ⅲ、Ⅳ级胶质瘤标本17例中强阳性4例、阳性7例、弱阳4例、阴性2例,阳性表达率为88.24%(15/17),LI值为(30.06±22.94)%,Ⅰ、Ⅱ级与Ⅲ、Ⅳ级间差异显著,P<0.05。在脑胶质瘤中MMP-2mRNA主要表达于瘤细胞及血管基底膜的内皮细胞的胞浆,其表达是在胞浆中出现棕色或棕黄色染色。在Ⅰ、Ⅱ级胶质瘤标本23例中阳性3例、弱阳3例、阴性17例,阳性表达率为26.09%(6/23),LI值为(6.43±5.32)%,在Ⅲ、Ⅳ级胶质瘤标本17例中强阳性4例、阳性7例、弱阳1例、阴性5例,阳性表达率为70.59%(12/17),LI值为(29.88±23.96)%,Ⅰ、Ⅱ级与Ⅲ、Ⅳ级间差异显著,P<0.05。MMP-2蛋白及mRNA的表达在Ⅰ、Ⅱ级胶质瘤组明显低于Ⅲ、Ⅳ级胶质瘤组,与胶质瘤的恶性程度及进展呈正比,更有利于对人脑胶质瘤的恶性生物学行为进行评价和判断。
The objectives of this study were to investigate the expression of MMP-2 protein and mRNA in glioma cells and its signifi- cance in the occurrence and malignant progression of gliomas. Immunohistochemical method and in situ hybridization were adopted to detect the expression of MMP-2mRNA protein and mRNA in 5 cases of normal brain tissue and 40 cases of gliomas at different grades. The expression of both MMP-2mRNA protein and mRNA in normal brain tissue was negative. In the cases of brain gliomas, the expression MMP-2 protein mainly showed in the cytoplasm of the tumor cells and endothelial cells of vascular basement membrane,which could be detected through brown or yellow-brown staining in the cytoplasm. In 23 cases of grade I and II glioma specimens, positive expression was found in 4 cases,weak positive in 6 cases, negative in 13 cases, with a positive expression rate at 43.48% ( 10/23 ) and LI value at (5.91 ± 4. 78) % ; in 17 cases of grade III and IV glioma specimens, strongly positive expression was detected in 4 cases, positive in 7 cases, weak positive in 4 cases and negative in 2 cases, with a positive expression rate at 88. 24% (15/17) and LI value at (30. 06 ±22. 94) %. It can be seen that the difference between grade I,II and grade III,IV was significant ( P 〈 0. 05). In addition, the expression of MMP-2mRNA mainly appeared in the cytoplasm of tumor cells and endothelial cells of vascular basement membrane in brain gliomas ,which was detected through brown or yellow-brown staining in the cytoplasm. In 23 cases of grade I and II glioma specimens, positive expression was found in 3 cases, weak positive in 3 cases, negative in 17 cases,with a positive expression rate at 26.09% (6/23) and LI value at (6.43 +5.32) % ; in 17 cases of grade III and IV glioma specimens, strongly positive expression was detected in 4 cases, positive in 7 cases, weak positive in 1 cases and negative in 5 cases, with a positive expression rate at 70. 59% (12/17) and LI value at (29. 88± 23.96)%. This also shows a significant difference between grade I,II and grade III, IV with P 〈 0. 05. The expression of MMP-2 protein and mRNA in grade I and II case group of gliomas was significantly lower than that in grade III and IV case group. It shows a positive correlation with the malignancy and progression of gliomas, which is more conducive to the evaluation and judgment on the malignant biological behavior of human brain gliomas.
