摘要
目的利用结核分支杆菌噬菌体裂解法检测结核杆菌对利福平、异烟肼、链霉素敏感性。方法选用传统方法判定为利福平、异烟肼、链霉素的耐药或敏感菌株,采用噬菌体裂解法检测其对以上三种药物的敏感性,对比其特异性和敏感度。结果运用噬菌体裂解法检测结果显示,异烟肼药物浓度为1μg/ml时,其灵敏度为87.0%,特异度为93.3%,与传统方法的符合率为92.0%;链霉素药物浓度为4μg/ml时,其灵敏度为96.2%,特异度为92.0%,与传统方法的符合率为92.9%;利福平药物浓度为5μg/ml时,其灵敏度为91.4%,特异度为96.2%,与传统方法的符合率为94.7%。结论采用噬菌体裂解法检测结核杆菌药物敏感性简便易行,灵敏度、特异度高,与传统方法比较符合率较高,便于临床推广。
Objective To detect mycobacterium tuberculosis sensitivity to rifampiein, isoniazid and streptomycin with phage splitting method. Methods We compared the specificity and sensitivity of the sensitive or resistant strains of rifampicin,isoniazid and streptomycin using the traditional method and phage splitting method. Results The detec- tion results using phage splitting method for display,drug concentration of isoniazid, streptomycin and rifampicin was 1μg/ml,4 μg/ml and 5 μg/ml ,the sensitivity was 87.0% ,96.2% and 91.4% ,the specificity was 93.3% ,92.0% and 96.2% ,the coincidence rate compared with the traditional method was 92.0% ,92.9% and 94.7 ,separately. Conclu- sion Detection drug sensitivity of mycobacterium tuberculosis using phage splitting method is simple,high sensitivity, high specificity,high coincidence rate compared with the traditional method and convenient clinic.
