软坚护肝片对大鼠肝纤维化的治疗作用 被引量:8

摘要 目的探讨软坚护肝片对四氯化碳(CCl4)诱导的大鼠肝纤维化的治疗作用及其可能的作用机制。方法将90只SD大鼠随机分为正常对照组、模型组、扶正化瘀胶囊阳性对照组和软坚护肝片1.24、0.62和0.31g/kg剂量组,除正常对照组外,其余各组采用CCl4灌胃建立大鼠肝纤维化模型共12周,于第9周治疗组每天灌喂软坚护肝片(1.24、0.62和0.31g/kg)和扶正化瘀胶囊(0.20 g/kg)干预8周。实验结束后检测大鼠血清丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、白蛋白(Alb)、透明质酸(HA)和Ⅲ型前胶原酶(PCⅢ)的水平,以及肝组织中羟脯氨酸(Hyp)的含量和肝指数。HE染色观察各组大鼠肝纤维化程度。结果与模型组比较,各剂量的软坚护肝片及扶正化瘀胶囊能显著降低大鼠血清中ALT、AST、HA和PCⅢ的水平,降低肝组织中异常升高的Hyp含量(P<0.05);同时,各剂量的软坚护肝片能显著提高大鼠血清中的Alb含量(P<0.05)。软坚护肝片各剂量组大鼠肝脏纤维化程度明显减轻。结论软坚护肝片有一定的抗肝纤维化作用,其作用机制可能与降低胶原的合成及抗自由基有关。 Objective To investigate the therapeutic effect of Ruanjian Hugan tablet on hepatic fibrosis induced by carbon tetraehloride( CC14 ) in rats and the possible mechanism. Methods Ninety SD rats were randomly divided into normal control group, model group, Fuzheng Huayu capsule group, Ruanjian Hugan tablet 1.24, 0. 62 and 0. 31 g/kg groups. In order to prepare the hepatic fibrosis model, all the rats except those of the normal control group were intragastrically administrated with CC14for twelve weeks. From the ninth week the treatment groups were given different doses of Ruanjian Hugan tablet and Fuzheng Huayu capsule for eight weeks. Blood samples were collected to determine the serum concentration of alanine aminotransferase ( ALT), aspartate aminotransferase (AST), albumin(Alb), hyaluronic acid (HA) and proeollagen-]lI (PC llI ). The liver was taken to measure the quantity of the hydroxyproline (Hyp) and calculate the liver index. Histopathologieal changes of the liver were evaluated by hematoxylineosin (HE) stain. Resdts Compared with the model group, the serum concentration of AST, ALT, HA, PC m and the levels of Hyp in liver in the treatment groups decreased significantly, while the serum concentration of Alb increased(P 〈 0.05 ). The HE stain results showed that the hepatic fibrosis in the rats was mitigated in Ruanjian Hugan tablet treated groups. Condusion Oral administration of Ruan- jian Hugan tablet has an anti-bepatic fibrosis effect in the rats induced by CCI4. The protective effect against hepatic fibrosis may be due to its ability to reduce the synthesis of collagen and block the effect of free radicals generated by CCl4.
