摘要
本实验室从果园土壤中筛选到一株产谷胱甘肽的酵母菌YS10。经26SrDNA鉴定为酿酒酵母(Saccharomyces cerevisiae),正交试验获得该菌株产谷胱甘肽合适的发酵培养基为:葡萄糖为3.0%、酵母膏为0.7%、硫酸铵为1.0%、磷酸二氢钾为0.3%、硫酸镁为0.05%。通过发酵条件的优化,该菌在500mL三角瓶装量50mL,30℃摇瓶培养72h,谷胱甘肽的产量可达183.04mg/L,是一株具有工业化开发潜力的谷胱甘肽生产菌株。
A Glutathione high-yield YS10 strain was screened from the soil of a orchard, which turned out to be a Saccharomyces cerevisiae according to the 26SrDNA analysis. The compositions of culture medium for the growing of YSI0 was obtained by orthogonal tests, which are 3.0% of glucose, 0.7% of yeast extract, 1.0% of ammonium sul- fate, 0.3% of potassium dihydrogen phosphate, 0.05% of magnesium sulfate.The result of this research shows that the opti- mal conditions for fermentation for YS10 strain was liquid volume of 50ml in 500mL erlenmeyer flask for 72h. The optimal cultivation temperature was 30℃. Under the optimal conditions the Glutathione yield was 183.04mg/L, which turned out to be a potential glutathione-produetion strain for its industrialization.
