高温与多聚甲醛方法杀灭结核杆菌效果的观察 被引量:3

摘要 目的观察高温与多聚甲醛方法杀灭结核杆菌的效果。方法参照消毒灭菌方法的原理,对20例临床分离的结核菌株分别进行高温、多聚甲醛处理,再将处理后或未处理的菌悬液接种至7H10培养基,置于37℃和5%CO2的环境培养8周并观察其生长情况。鉴于可疑生长的菌株,实行第二次接种培养观察。结果基于对照组20例菌株生长均良好的情况,多聚甲醛处理后的菌株均不生长,而65、70、85℃高温处理后的菌株分别有1、2、4例菌株不生长,其余菌株在培养至7~10天时形成针尖样的可疑微小菌落,但经第二次接种与培养后证实可疑菌株均不生长。结论高温与多聚甲醛处理方法均可快速杀灭结核杆菌,其效能是稳定、可靠的。 [ Abstract ] Objective To observe the effect of killing mycobacterium tuberculosis using high temperature and paraformaldehyded. Methods Based on the principle of disinfection and sterilization, 20 clinical isolates of my- cobacterium tuberculosis were exposed to high temperature or paraformaldehyded. Following different treatments, sus- pension of each M. tuberculosis isolate was inoculated to 7H10 medium, and observed the growth state after cultiva- tion of 8 weeks at 37℃ in an environment of 5% CO2. In view of the doubtful M. tuberculosis isolates, they were re- ceived the second inoculation and cultivation. Results Based on well growth status of the room temperature controls of 20 clinical isolates, there was obviously no growth among M. tuberculosis with paraformaldehyded treatment, and there was obviously 1, 2, 4 isolate with no growth after high temperature (65℃ ,70℃ ,85℃ ) treatment, respective- ly, the rest can doubtfully shape needlepoint - like tiny bacterial colonies after cultivation of 7 - 10 days, which had been confirmed to deactivate through the second inoculation and cultivation. Conclusion The efficacy was stable and reliable to kill M. tuberculosis quickly using high temperature or paraformaldehyded method.
