摘要
目的探讨CGRP对血管内皮细胞和成骨细胞共培养体系成骨增殖和钙化的作用。方法建立成骨细胞和血管内皮细胞共培养体系,采用含有10-7、10-8、10-9、10-10mol/LCGRP和无CGRP的培养液培养,对各浓度CGRP对于各组的成骨增殖和钙化进行检测。结果通过茜素红染色发现在培养液中不添加CGRP组钙化结节形成明显最多,添加10-7、10-8、10-9、10-10mol/LCGRP各组钙化结节形成与CGRP浓度近似于相反的变化趋势。通过检测细胞增殖发现CGRP对于共培养系统中的成骨细胞增殖具有促进作用。结论 (1)CGRP对于成骨细胞增殖具有促进作用,从测试中所采用的各个浓度来看,10-9mol/LCGRP刺激成骨细胞增殖较明显。(2)高浓度CGRP对于钙化可能存在抑制作用。
Objective To explore the effect of calcitonin gene-related peptide (CGRP) on the proliferation and calcification of the co-culture system of vascular endothelial cells and osteoblasts. Methods The established co-culture system was cultured by the culture medium with CGRP at different concentration of 10^-7、10^-8、10^-9、10^-10mol/L. and the medium with no CGRP; the proliferation and calcification were detected. Results The Alizarin Red staining revealed that more calcified nodules were formed when cultured by the culture medium with no CGRP while less nodules were found when cultured by the culture medium with CGRP at different concentration of 10^-7、10^-8、10^-9、10^-10mol/L. Conclusions (1) CGRP can promote the osteoblasts proliferation and CGRP of 10^-9mol/L is obviously effective in stimulating the osteoblasts proliferation; (2) CGRP of high concentration may inhibit calcification.
