东北雷公藤试管苗培养的研究

摘要 为保护野生资源和实现栽培,该研究以具有腋芽的嫩茎为材料,采用组织培养的方法,进行了腋芽的生长培养、生长芽生根培养、试管苗的生根继代繁殖培养、试管苗的移栽和定植的研究。结果证明:MS+蔗糖30g.L-1+GA30.4mg.L-1+ZT 0.1mg.L-1+NAA 0.05mg.L-1是腋芽生长培养的适宜培养基;将从基部剪下的生长芽或试管苗茎段下部切口在10mg.L-1的IAA溶液中蘸一下、接种到1/2MS+ABA2号0.5mg.L-1+蔗糖10g.L-1+IBA0.1mg.L-1培养基上进行生根培养,是生长芽生根培养和生根继代繁殖培养的理想方法。试管苗的移栽成活率为99%,定植成活率为99.3%。定植的试管苗不仅生长旺盛而且保持了东北雷公藤的所有植物学性状。 In order to protect wild resources and realize artificial cultivation, with the method of tissue culture, the tender stems of Tripterygirnu regeli were used as materials to do the research on growth of lateral buds, rooting of grow buds, roo ting multiplication of tube seedlings, transplantation and stable planting of tube seedlings. The results showed that MS+ su- crose30 g·L 1 +GAa0.4 mg·L-1 +ZT0.1 mg- L-1 +NAA 0.05 mg. L-1 is the suitable medium for lateral buds' growth; to cut the grow buds from the base or lower part of tube seedlings' stem and dip the section incision in the solution with IAA 10 mg - L-1 , then vaccination to the medium 1/2 MS+ABA2 0.5 mg·L-1 +sucrose 10 g·L-1 +IBA0. 1 mg·L -1 for rooting culture,it is a ideal method for rooting cultivation of grow buds and rooting multiplication of tube seedlings. The survival rate of tube seedlings transplanting is 99% and the survival rate of planting is 99.3%. The planted tube seedlings not only grow well and maintain all botany traits of Tripterygirnu regeli.
