解钾细菌的筛选、鉴定以及高效培养 被引量:15

Isolation,identification and high-efficiency cultivation of potassiumreleasing bacteria

摘要 从烟草根际土壤中成功筛选出10株解钾细菌,并选择其中四种透明圈明显且生长量较大的菌种进行了解钾活性测定,其中解钾细菌GJ07的发酵液中K^+离子质量浓度高出空白对照组54.5%.对GJ07进行16S rDNA的鉴定,确定其为巨大芽孢杆菌.采用批量发酵培养,通过改变碳源、氮源、碳氮比、温度、pH值、装液量等研究了解钾细菌GJ07生长的优化控制条件.结果表明蔗糖和蛋白胨为GJ07生长的最佳碳源和氮源(碳氮质量比为10:1),最适宜生长温度为35℃,最适宜初始pH值为7.00,最佳装液量为25 mL. Ten strains of potassium-releasing bacteria were isolated from the tobacco rhizosphere soil. Four strains with an obvious transparent zone and high efficiency proliferation were selected to investigate their potassium-releasing capacity. The K+ mass concentration in the fermentation broth of the strain G J07 is 54.5% higher than that of the blank control group. The strain G J07 was identified as Bacillus megaterium by the identification of 16S rDNA analysis. The batch culture conditions of the strain G J07 were optimized via adjusting carbon sources, nitrogen sources~ carbon- to-nitrogen mass ratio, temperature, pH values and loading liquid. It is found that sucrose is the best carbon source, peptone is the optimal nitrogen source, the optimal carbon-to-nitrogen mass ratio is 10:1, the best temperature and initial pH value are 35 ℃ and 7.00 respectively, and the optimal loading liquid is 25 mL.
