酵母蛋白tra1突变对NuA4复合体结合至核糖体RNA启动子效率的影响

摘要 核糖体是细胞合成蛋白的工厂,它的合成速率和数量与细胞的生长、分化、细胞寿命、胚胎发育、癌症发生有密切的关系。因此研究核糖体生物合成的分子机制具有非常重要的意义。之前实验通过Tra1的突变,发现Tra1蛋白结合至核糖体RNA基因启动子的效率大大降低。而Tra1蛋白是酵母细胞中的染色质重塑复合体SAGA和NuA4唯一共有的亚基,并且通过NuA4和SAGA复合体参与了核糖蛋白基因的转录。所以本实验以NuA4的Esa1和Epl1亚基为靶抗原,通过染色质免疫沉淀技术,对比野生株和突变株的核糖体RNA基因启动子与NuA4的结合效率,分析tra1突变对NuA4复合体结合至核糖体RNA启动子效率的影响。从而进一步为"研究Tra1的突变是通过什么途径影响核糖体RNA基因的表达效率"提供研究基础。
