摘要
目的建立HPLC同时测定怀槐树皮药材中染料木苷、鸢尾苷、saikoisoflavonoside A和怀槐异黄酮苷含量的方法。方法色谱柱为Diamonsil^(TM)ODS(250 mm×4.6 mm,5μm);流动相为乙腈-质量分数为0.05%磷酸溶液(体积比为16:84);流速为1.0 mL·min^(-1);检测波长为260 nm;柱温为35℃。结果染料木苷、鸢尾苷、saikoisoflavonoside A和怀槐异黄酮苷的质量浓度分别在0.42~2.09、1.00~5.00、5.12~25.60和1.81~9.04 mg·L^(-1)内与峰面积线性关系良好,平均回收率分别为99.4%、99.9%、100.2%和99.9%,RSD分别为2.1%、3.0%、2.5%和2.7%。结论该方法可用于怀槐树皮中染料木苷、鸢尾苷、saikoisoflavonoside A和怀槐异黄酮苷的含量测定。
Objective To develop a method for the simultaneous quantitative analysis of genestin, tectoridin, saikoisoflavonoside A and maackiaisoflavonoside of the stem bark of Maackia amurensis. Methods A Dia- monsilTM C18 column was used for HPLC;the mobile phase was acetonitrile and aqueous solution containing 0.05% (φ) phosphoric acid with the volume ratio of 16:84 and the UV detection wavelength was set at 260 nm;the flow rate was 1.0 mL-min-1 ;the oven temperature was 35 ℃. Results The linear ranges of genes- tin, tectoridin, saikoisoflavonoside A and maackiaisoflavonoside were 0. 42- 2.09,1.00- 5.00,5.12- 25.60 and 1.81- 9.04 mg· L - 1, respectively. The average recoveries ( n = 6 ) of genestin, tectoridin, saikoisoflavono- side A and maackiaisoflavonoside were 99. 4% with RSD of 2. 1,99.9% with RSD of 3.0,100. 2% with RSD of 2. 5,99. 9% with RSD of 2.7 ,respectively. Conclusions The method can be used to determine the four isoflavone glycosides in the stem bark of Maackia amurensis.
