摘要
目的:观察Von Hippel-Lindau(VHL)及Survivin在胃癌中的表达及相互关系,探讨联合检测VHL及Survivin基因在胃癌中的意义。方法:采用免疫组织化学SP法检测40例胃癌组织,30例正常胃黏膜组织中VHL及Survivin基因的表达。结果:40例胃癌组织中,VHL基因的阳性表达率为35%,显著低于正常胃组织的表达(P<0.01),Survivin的阳性表达率为80%,显著高于正常胃组织的表达(P<0.01),VHL基因的表达与胃癌的临床分期(P<0.05)、淋巴结转移(P<0.01)有关,Survivin基因的表达与胃癌的临床分期(P<0.01)、淋巴结转移(P<0.01)有关。结论:VHL及Survivin基因的表达可能与胃癌的发生发展有关,VHL基因蛋白的表达缺失可能在胃癌的发生、发展中存在重要意义,VHL可能作为诊断胃癌新的参考指标。
Objective. To study the expressions of Von Hippel--Lindau（VHL）and Survivin in gastric carci- noma, and the correlation between VHL and Survivin expression in gastric carcinoma tissue. Methods： The ex- pressions of VHL and Survivin were detected in 40 cases of surgically resected gastric carcinoma specimens and 30 cases of corresponding normal mucosa tissues using the immunohistochemistry S--P method. Results： In 40 cases of gastric carcinoma tissue, the positive expression rates of VHL were 35 %, which was lower than that in normal gastric mucosa tissues （P 〈 0.01）, the positive expression rates of Survivin were 80%, which was higher than that in normal gastric mucosa tissues （P 〈 0.01）. The expression of VHL gene significantly corre- lated with TNM stages （P 〈 0. 05）, lymph node metastases （P〈0.01）. The expression of Survivin gene sig- nificantly correlated with grade of cell differentiation, TNM stages （P 〈0.01）, lymph node metastases （P〈 0.01）. Conclusion： The expressions of Von Hippel--Lindau （VHL）and Survivin in gastric carcinoma maybe correlate with the occurrence and development of gastric carcinoma. Loss of VHL gene expression may be of great significance in occurrence and development of gastric carcinoma. VHL may be used as a new index for the diagnosis of gastric carcinoma.
