大鼠体内兰索拉唑血药浓度测定方法的研究

Study on HPLC Determination Method of Blood Drug Level of Lansoprazole in Rat body

本文建立大鼠血浆测定的HPLC法,研究大鼠血浆中兰索拉唑的药动学。应用HPLC法,替硝唑为内标,用C18柱分离,用乙醚-二氯甲烷(3:2)提取处理,流动相:乙腈-水(36:64,0.2%三乙胺)磷酸调pH9.0,流速0.7mL.min-1。兰索拉唑在大鼠血浆中线性为20.0～20000ng.mL-1,日内、日间RSD均小于15%。本方法专属、稳定。

Establish a HPLC method for determination of lansoprazole in rat plasma,to study the pharmacokinetic of lansoprazole in rat plasma.Tinidazole was used as internal standard and the samples were extracted by ether-dichloromethane（3：2）,The separation was carried out on a C18 column using a mobile phase consisting of 0.2%triethylamine acetonitrile-water（36∶64）pH9.0 at 0.7mL·min-1.The linear calibration curves were obtained in the concentration range of 20.0~20000ng·mL-1 in rat plasma.The intra-and inter-run precisions were lower than 15% in terms of RSD.
