The expression and roles of TLR9 in esophageal cancer cell

目的观察Toll样受体9(TLR9)在食管鳞癌细胞的表达及作用。方法用RT-PCR方法检测食管鳞癌细胞系TE10、TE30和TE70上TLR9的表达;流式细胞术检测TE10细胞上TLR9的表达;用TLR9的激动剂CpG寡脱氧核苷酸(CpG ODN)10μg/ml分别作用TE10细胞12 h、24 h和48 h后,用MTT法检测细胞生长抑制率。结果 TE10细胞上有TLR9的表达;TLR 9激动剂CpG ODN作用于TE10细胞后不同时间均能明显促进TE10细胞生长(P<0.01)。结论 TLR9激动剂CpG ODN可促进食管鳞癌细胞的增殖,提示TLR9可能是食管鳞癌治疗的靶点之一。

Objective To investigate the expression and possible mechanism of TLR9 in the incidence of human esophageal squa- mous cell carcinoma cells. Methods The expression of TLR9 in esophageal squamous cell carcinoma cell lines TE10, TE30 and TE70 cells was measured by RT-PCR and the expression of TLR9 in TE10 was detected by flow cytometry as well. The inhibitive effect of 10 μg/ml TLR9 agonist （ CpG ODN） in TE10 cells at 24 h, 48 h and 72 h was observed by MTr assay. Results TLR9 was expressed in TE10 cells. TLR9 agonist CpG ODN promoted the growth of TE10 cells （P 〈 0, 01 ） at different time point. Conclusions TLR9 agonist CpG ODN could promote the proliferation of esophageal carcinoma cells indicating that TLR9 may be one of the targets of squamous cell carcinoma treatment.
