摘要
目的观察当归挥发油对实验性哮喘大鼠的平喘作用及其对IL-4、IFN-γ的影响。方法采用注射OVA与氢氧化铝致敏、OVA雾化吸入激发来复制哮喘动物模型,通过观测特异性与非特异性支气管激发实验中大鼠的行为、测定血清中IgE的含量与BALF中IL-4和IFN-γ的水平,探讨当归挥发油的平喘作用及其作用机制。结果当归挥发油能促进大鼠体质量的增加,明显缓解OVA或乙酰胆碱与组胺混合物激发实验中大鼠的哮喘等行为学表现,降低血清与BALF中IgE与IL-4水平,提高BALF中IFN-γ水平。结论当归挥发油具有一定的平喘、治疗支气管哮喘的作用,调节IL-4与IFN-γ的表达失衡是其作用机制之一。
To investigate anti-asthma effects of volatile oil of radix angelicae sinensis (VOA) and its possible mechanisms, a model of rat asthma was established with injecting OVA and AI(OH)3 for sensitization and administering OVA aerosol for challenge. Asthmatic behaviors of rat under specific or nonspeeific bronchial provocation had been evaluated and scored; the levels of total IgE (T-IgE) in serum, and the levels of IL-4 and IFN-γ in bronchoalveolar lavage fluid (BALF) had been quantified. Our results showed that VOA could increase body weight of rats, relieve rat's asthmatic behaviors after OVA or Acetylcholine + Histamine stimulus on bronchi,downgrade the levels of T-IgE in serum and IL-4 in BALF, but upgrade the concentration of IFN-γ in BALF. In conclusion, VOA has certain effects of anti-asthma and one of mechanisms is to recovering the equilibrium of IL-4 and IFN-γ.
