褪黑激素对增生性瘢痕成纤维细胞分泌转化生长因子β1的影响及其机制 被引量:2

摘要 目的 研究褪黑激素对增生性瘢痕Fb分泌TGF-β1的影响及机制。方法 采用组织块法分离培养人增生性瘢痕Fb,按随机数字表法将细胞分为褪黑激素组、褪黑激素+Luzindole(褪黑激素受体拮抗剂)组、Luzindole组和对照组(仅以细胞培养液培养),前3组每孔细胞分别添加相应物质与细胞培养液培养,褪黑激素终浓度为1×10-3 mmol/L、Luzindole终浓度为1 mmol/L。 每组10个复孔。培养24 h,采用ELISA法检测细胞培养上清液中TGF-β1含量,实时荧光定量PCR检测细胞TGF-β1、Smad 7和Rho激酶(ROCK)mRNA表达。对数据进行单因素方差分析或LSD-t检验或Kruskal-Wallis检验,组间比较采用Nemneyi法。结果 褪黑激素组Fb上清液中TGF-β1含量为(2.8±2.4)pg/mL,低于褪黑激素+Luzindole组的(23.9±2.7)pg/mL、对照组的(28.9±2.0)pg/mL和Luzindole组的(24.9±2.8)pg/mL(F=94.58,P〈0.05)。褪黑激素组细胞TGF-β1 mRNA表达量为11.9±1.2,明显低于其他组(F=20.79,P〈0.05);褪黑激素+Luzindole组TGF-β1 mRNA表达量(14.9±1.8)与对照组(17.2±0.6)比较差异无统计学意义(t=0.806,P〉0.05)。褪黑激素组Fb Smad 7 mRNA表达量(0.076±0.080)明显高于对照组(0.023±0.024)、褪黑激素+Luzindole组(0.026±0.026)、Luzindole组(0.037±0.021),χ2=9.567,P〈0.05。褪黑激素组ROCK mRNA的表达量明显低于对照组(0.002 30±0.002 70)、褪黑激素+Luzindole组(0.001 30±0.001 20)和Luzindole组(0.001 10±0.001 20), χ2=9.420,P〈0.05。结论 褪黑激素可通过与受体结合,上调瘢痕Fb中Smad 7表达、下调ROCK表达,从而抑制增生性瘢痕Fb表达TGF-β1。
