摘要
目的研究Nod样受体热蛋白结构域相关蛋白3(NLRP3)在人脑胶质瘤组织中的表达,探讨NLRP3在人脑胶质瘤的发生、发展中的作用。方法按照2007年WHO中枢神经系统肿瘤分类及分级标准,将所收集的49例人脑胶质瘤组织根据其病理类型分为低级别组(20例)和高级别组(29例),并收集脑外伤后行颅内减压手术切除的正常脑组织10例做对照。应用免疫组织化学技术,并采用实时荧光定量逆转录聚合酶链反应(QRT-PCR)和免疫印迹技术,检测NLRP3在各组人脑胶质瘤及人正常脑组织中的表达。结果 NLRP3在正常脑组织中表达不明显,在低级别胶质瘤中表达较少,在高级别胶质瘤中表达明显。NLRP3 mRNA在正常脑组织(1.04±0.52)、低级别组(1.85±1.02)和高级别组(3.36±1.38)比较,差异有统计学意义(P<0.01)。人脑胶质瘤组织中NLRP3在mRNA水平及蛋白水平均高于正常脑组织(P<0.01)。NLRP3的表达与人脑胶质瘤的恶性程度呈正相关。结论人脑胶质瘤中存在NLRP3的异常表达,其表达量随胶质瘤恶性程度的升高而增加,NLRP3在人胶质瘤的发生和侵袭中可能发挥着重要的作用,检测NLRP3的表达有助于胶质瘤分化程度的判断。
Objective To research the expression of Nod-like receptor pyrin domain-containing protein 3 ( NLRP3 ) in glioma, and to investigate the role of NLRP3 in glioma genesis and improvement. Methods 49 cases of human glioma were graded according to World Health Organization (WHO) 2007 criteria, and were divided into the low grade group (WHO I-lI , 20 cases) and the high grade group( WHO m-IV, 29 cases). The expression of NLRP3 was measured by immunohistochemistry in 49 human glioma tissues and 10 normal brain tissues. The expressions of NLRP3 mRNA and protein were detected by real-time PCR and Western blot. Results The NLRP3 expression was negative in 10 normal brain tissues and low in the low grade group. There was obvious expression of NLRP3 in the high grade glioma group. The level of NLRP3 mRNA and protein in normal group, low grade group and high grade group was 1.04 ~ 0.52, 1.85 _+ 1.02 and 3.36 _+ 1.38 respectively. There were significant differences among the three groups(P 〈0.01 ). Com- pared with normal brain tissue, the expression of NLRP3 in human glioma in both mRNA and protein levels were higher ( P 〈 0.01 ). Conclusion There is abnormal expression of NLRP3 in glioma, and the NLRP3 level is positively corre- lated with malignancy of human glioma. NLRP3 may play an important role in the genesis and invasion of glioma. The expression of NLRP3 can be used to estimate the malignancy of glioma.
