摘要
以甘草无菌苗茎尖为材料,采用5因素4水平正交试验,以诱导率、不定芽数、叶片数、株高和鲜重为指标,研究6-BA、KT、NAA、蔗糖和不同培养基对甘草无菌苗茎尖诱导的影响,并采取排队评分与公式评分法对几个指标打分后进行分析,优化出最佳的激素配比,建立甘草茎尖快繁体系。结果表明:不同培养基对甘草茎尖诱导苗的影响最大,6-BA对甘草茎尖诱导苗的生长发育影响次之,蔗糖对甘草茎尖诱导苗生长发育影响最小,甘草茎尖诱导苗最佳激素配比为:MS+6-BA 0.3mg/L+NAA 1.0mg/L+KT 0.3mg/L+蔗糖30g/L;增殖培养基MS+6-BA1.0mg/L+NAA0.1mg/L+蔗糖30g/L,增殖倍数为7倍,平均株高为5.4cm;生根培养基MS+KH2PO413.5mg/L+蔗糖30g/L,生根率达到100%。
This experiment used the material of GZycyrrhiza uralensis Fisch. shoot-tips and took the method of orthogonal experiment of 5 factors and 4 levels to research the 6-BA.KT. NAA, sucrose and different medium's induced influence on germfree plants shoot-tip of Glycyr- rhiza. Used line score and formula scoring methods to analyse several index after grading, opti- mized the best ratio of hormone to establish the rapid propagation system of shoot-tips. Results showed that culture mediums had the greatest impact, 6-BA was the minor, Sucrose had the least impact. The suitable hormone proportion of culture medium for Glycyrrhiza shoot-tip was MS+6-BA0.3 mg/L+NAA1.0mg/L+KT0.3 mg/L+sucrose 30 g/L. The suitable consists of proliferation medium was MS+6--BA1.0 mg/L+NAA0.1 mg/L+sucrose30 g/L,prolifer- ation ratio reached 7times, average plant height was 5.4 cm;The suitable consists of rooting medium was MS+KH2PO413.5 mg/L+sucrose 30 g/L and rooting rate reached 100%.
