摘要
本试验利用猪轮状病毒和大肠杆菌混合感染建立腹泻模型,通过免疫组织化学方法研究白头翁素对腹泻小鼠肠黏膜修复因子蛋白表达的影响,揭示白头翁素防治腹泻的黏膜修复机制。结果显示:小鼠腹泻96h内,自愈组、治疗组和预防组黏膜修复因子PCNA和TGFα的蛋白表达量均有所升高,其中预防组显著高于空白对照组(P<0.05),腹泻96h后,表达量逐渐下降直至正常。结果表明,白头翁素防治小鼠轮状病毒和大肠杆菌混合感染性腹泻的显著效果可能与其调节黏膜修复因子PCNA和TGFα的表达有关,从而促进了肠黏膜的修复。
Nemonin is a main component of pulsatilla, which is extensively used to control ani- mal diarrhoea. However, the mechanism underlying how anemonin cures diarrhea is not fully clear. To investigate the intestinal mucosa repair mechanism of anemonin on diarrhea, the ex- periment established a diarrhoea model by PRV and E. coli by using the method of immunohis- tochemistry. The results showed that, within the diarrheal stage of 96 hours, the protein ex- pression of PCNA and TGFa in the self-healing group, treatment group and prevention group was all higher than that in control group, and the expression in the prevention group was sig- nificantly higher than that in control group (P〈0.05). After the stage of 96 hours, the expression of PCNA and TGFa gradually decreased to normal condition. The results suggested that anemonin could significantly prevent diarrhoea caused by PRV and E. coli due to the expression of PCNA and TGFa.
