摘要
不同的基本培养基对枇杷胚愈伤组织的诱导有较大影响 ;不同品种的鱼雷形胚的愈伤组织的诱导率有明显差异 ;不同生长调节剂组合和碳源对愈伤组织的形成有较大的影响 .比较而言 ,以 B5为基本培养基并添加 0 .5mg· L-12 ,4 - D和 1mg· L-1BA组合诱导的愈伤组织的质量最佳 .蔗糖作为碳源 ,有利于愈伤组织生长 ,而山梨醇则对细胞分化有利 .附加 0 .1mg· L-12 ,4 - D和 0 .5mg· L-1BA的 B5培养基有利于胚性愈伤组织的继代、增殖 ,低温和弱光照有利于胚性愈伤组织的保存 .
There are significant effects of different basal media, growth regulator combinations and carbon sources on induction and formation of loquat callus, and there is obvious difference in induction rates of torpedo shaped embryo calli among various varieties. Comparatively, B 5, the basal medium, added with 0.5 mg·L -1 2,4 D and 1 mg·L -1 BA is able to induce the callus with optimal quality. Sucrose, the carbon source, is helpful to callus growth while sorbitol beneficial for cell differentiation. B 5 added with 0.1 mg·L -1 2,4 D and 0.5 mg·L -1 BA promotes subculture and regeneration of embryogenic callus, meanwhile low temperature and weak illuminance are favourable to its preservation.
