隔指孢属rDNA ITS区间限制性片断长度多态性(RFLP)分析 被引量:2

摘要 利用PCR-RFLP方法对捕食线虫真菌隔指孢属进行了系统发育研究。以 ITS1和ITS4为引物对该属8种8个菌株的核糖体DNA转录间区(ITS)进行了 PCR扩增, 4种内切酶( AluⅠ、 Hae Ⅲ、 Hpa Ⅱ、 TaqⅠ)酶切。结果表明该属的 ITS区长度没有明显差异,其长度范围在564~595之间,酶切图谱能体现不同种 间的多态性。根据4种内切酶酶切结果,利用UPGMA法构建的隔指孢属分子系 统树,基本体现了属内不同种间的亲缘关系,从分子水平证明了隔指孢属形态分类 上的合理性。 The systematic relationships of 8 strains of Dactylella representing 8 species were evaluated by using PCRRFLP analysis. ITS was amplified and digested with four base restriction enzymes (Alu Ⅰ, HaeⅢ, Hpa Ⅱ, Taq Ⅰ ). The results showed that the length of ITS varied among isolates ranging between 564 and 595 bp. The restriction digestion patterns were distinctly different among species. Systematic relationshops was embodied basically by molecular phylogenetic tree of 8 strains based on the production of 4 base restriction enzymes digesting by using UPGMA. The result suggested that morphological classification of Dactylella is reasonable.
