The effect of pre-treatment of frozen-thawed semen with caffeine on in vitro fertilization of bovine oocytes

摘要 用 2.5mM咖啡因处理解冻牛精液 10min.或 5.0mM咖啡因处理 30min, 精子经离心后加入到不含咖啡因的受精液中进行体外受精,均获得较高卵裂率(分 别为 89. 1%、 89. 9%) 及囊胚率(分别为 44. 4%、 45. 9%),并且第 7 天一级囊胚 数较多。 This experiment was designed to eliminate caffeine in FM by pre-incubation of semen with different concentrations of caffeine at different period of time. According to the results, when semen being pre- incubated with 2. 5mM caffcine for 10 minutes or 5 .0 mM caffeine for 30 minutes and put into FM without caffeine, high cleavage rates (89. 1 %, 98.9 %, respectively) and blastocycst rates (44 .4 %, 45. 9% respectively) can be achieved and were significantly higher than that of group 10, both pre--treatments also gave much more grade one blastocycst on the seventh day which stand for high quality. Comparing with control group, no significant difference existed in cleavage rate and blastocycst rate, but positive tendency was showed.
