摘要
目的:应用彩色超声心动图观察益心泰对慢性心衰大鼠心肌重构的影响。方法:用冠脉结扎法造模,将造模成功的50只健康Wistar雄性大鼠分为5组:模型组,益心泰高、中、低剂量组,培哚普利组,每组10只,于造模成功后第50天开始给药。益心泰高、中、低剂量组均灌服益心泰丸,剂量分别为10.5g.kg-1.d-1、5.5g.kg-1.d-1、2.5g.kg-1.d-1;培哚普利组灌服4.5g.kg-1.d-1;模型组灌服同培哚普利组同等容积的生量盐水。均每天1次,共用药8周。另设正常对照组10只,对照组不进行药物干预。用药8周后心脏超声检测室间隔厚度(IVS)、左心室收缩期后壁厚度(LVPWs)、左心室舒张期后壁厚度(LVPWd)、左心室舒张末内径(LVEDD)、左心室收缩末内径(LVESD)、射血分数(EF)。结果:与模型组比较,益心泰高、中、低剂量组和培哚普利组LVEDD、LVESD均显著降低,LVPWs、LVPWd、EF、IVS明显升高,差异有统计学意义(P<0.05)。益心泰高剂量组与培哚普利组各项指标比较,差异无统计学意义(P>0.05)。结论:益心泰确能改善慢性心衰大鼠心肌重构,提高心功能。
Objective:To observe the effect of Yixintai on the myocardial remodeling in rats with chronic heart failure by color Doppler echocardiography. Methods: A rat model of chronic heart failure was induced by coronary artery ligationin all groups, and 50 successful model rats were equally divided into five groups : model group, high - dose, middle - dose, and low - dose Yixintai groups, perindopril group, and the rats began to receive their respective medications 50 d later. The high - dose, middle - dose, and low - dose Yixintai groups received Yixintai pills( 10. 5g.kg-1 .d-1 ,5.5g.kg-1 .d-1 ,and 2. 5g.kg-1. .d-1 )by gavage; the perindopril group re- ceived perindopril(4. 5g.kg-.1 d-1)by gavage; the model group received an equal volume of saline by gavage; The medication was given once daily in each treated group. In addition, we divided another 10 rats into normal control group, the normal control group re- ceived no medication. Interventricular septal thickness (IVS) ,left ventricular systolic posterior wall thickness (LVPWs) ,left ventricu- lar diastolic posterior wall thickness ( LVPWd), left ventricular end - diastolic diameter ( LVEDD), left ventricular end - systolic diam- eter (LVESD) , and ejection fraction (EF) were measured by eehocardiography after 8 weeks of medication. Results:Compared with the model group, the high - dose, middle - dose, and low - dose Yixintai groups and perindopril group had significantly decreased LVEDD and LVESD and significantly increased LVPWs, LVPWd, EF, and IVS ( P 〈 0. 05 ). There were no significant differences in the indices between the high - dose Yixintai group and perindopril group (P 〉 0. 05 ). Conclusion: Yixintai can attenuate myocardial remodeling and improve cardiac fimction in rats with chronic heart failure.
