核孔蛋白复合体蛋白88在乳腺癌细胞株MCF-7和MDA-MB-435S中的表达及意义研究 被引量:4

摘要 目的探讨核孔蛋白复合体蛋白88(NUP88)和mRNA在乳腺癌细胞株MCF-7和MDA-MB-435S中的表达及意义,验证NUP88可能是乳腺癌的重要肿瘤标记物,从而为临床开发新的乳腺癌靶向药物提供了新的靶标。方法采用Western blot检测乳腺癌细胞株MCF-7和MDA-MB-435S中NUP-88的表达;用反转录聚合酶链反应(RT-PCR)法检测MCF-7和MDA-MB-435S中NUP88 mRNA的含量。结果乳腺癌细胞株MDA-MB-435S中NUP88表达水平明显高于MCF-7,差异有统计学意义(P<0.01);NUP88表达水平与乳腺癌细胞株的侵袭性有关。NUP88 mRNA在乳腺癌细胞株MDA-MB-435S中的表达水平高于MCF-7,差异有统计学意义(P<0.01)。结论 NUP88及mRNA的表达与乳腺癌细胞株的侵袭性有关,提示NUP88将来有可能可作为判断乳腺癌恶性程度,预测侵袭转移和评估预后的参考指标。 Objective Research nuclear pore protein complex protein 88 (NUll8) protein and mRNA in breast cancer cell lines MCF -7 and MDA - MB -435S the expression and significance. NUP88 validation may be the important breast cancer tumor marker, thus far clinical development new breast cancer targeted drugs provided new targets. Methods Western blot detection of MCF-7 and MDA -MB -435S NUP -88 protein expression. With RT -PCR method to detect MCF -7 and MDA - MB -435S in NUP88 mRNA content. Results Breast cancer cell line MDA - MB -435S of NUP88 protein expression was significantly higher than that in MCF -7 (P 〈0. O1 ). The expression of NUP88 protein levels and breast cancer cell inva- siveness related. NUP88 mRNA in breast cancer cell lines MDA - MB -435S the expression level higher than breast cancer cell lines MCF - 7 (P 〈 0.01 ). Conclusion The NUP88 protein and mRNA expression and breast cancer cell lines involved in the invasion, suggesting that NUP88 likely in the future may be judged as to breast malignancy, metastasis and prognosis prediction of the reference index
