甲状腺功能减退大鼠肾脏形态及体视学研究 被引量:3

THE MORPHOLOGY AND STEREOLOGY STUDY OF KIDNEY IN HYPOTHYROID RATS

摘要 目的研究甲状腺功能减退症(甲减)大鼠肾脏形态及体视学变化,探讨甲状腺激素水平对肾脏的影响。方法 Wistar大鼠随机分为2组,甲减组和对照组,2组大鼠通过不同碘含量饮食复制甲减动物模型,进行肾脏形态定量学观察。结果与对照组比较,甲减组总甲状腺素、游离甲状腺素、总三碘甲状腺原氨酸、游离三碘甲状腺原氨酸均显著下降(P<0.01);大鼠体质量及肾质量明显减轻(P<0.05);肾小球、肾小管基底膜增厚;肾小球平均面积和平均体积著降低(P<0.05);肾小球内代表系膜基质含量的PAS阳性物质积分光密度值显著增高(P<0.01)。结论甲减可导致Wistar大鼠肾小球、肾小管基底膜增厚;肾小球萎缩、PAS阳性物质增多。 Objective To study the morphology and stereology changes of kidney in hypothyroid rat. To approach the effect of thyroid hormone to kidney. Methods Wistar rats were randomly divided into 2 groups as control group and hypothyroid group. The rats of hypothroid group were fed with different indine diet to duplicate hypothyroidism animal model. The morphometric parameters change of kidney were observed by routine histology method. Results Compared with the control group, serum free triiodothyronine, free thyroxine, total triiodothyronine and total thyroxine were markedly lower in hypothyroid group ( P 〈 0.01 ). The body weight and kidney weight diminished markedly ( P 〈 0.05 ). The basal membrane of glomerulus and renal tubule of periodic were markedly thicker. The mean glomerular area and volume diminished markedly ( P 〈 0.05 ). The integral optical density (IOD) value acid-shiff reaction (PAS) positive material which stand for the content of mesangial matrix was increased significantly( P 〈 0.01 ). Conclusion Hypothyroidism can lead to the thickening of glomerular and tubular basement membrane, glomerular atrophy, and increase of PAS-positive material in Wistar rats.
