摘要
目的建立脑胶质瘤标本库,充分保护和合理应用胶质瘤标本,为胶质瘤的临床及科研提供肿瘤标本。方法收集脑胶质瘤患者手术前静脉血、获取新鲜肿瘤组织标本和部分正常脑组织标本,分别提取血液及组织标本中DNA、RNA以及蛋白,贮存于-196℃液氮罐中,记录每例患者流行病学特点及病理诊断,术后6个月开始进行随访。结果全血及组织中提取的基因组DNA、组织标本中提取的总RNA其OD260/280比值都在1.8~2.1之间、所提取蛋白经二辛可酸法蛋白定量检测均能够满足胶质瘤分子生物学研究的需要。结论脑胶质瘤标本库的初步建立,形成了一套系统的、可行的胶质瘤标本库的建设及管理方法,可为胶质瘤的分子生物学研究提供标本和相关资料来源。
Objective To protect and utilize the tumour tissue resources fully and provide a suitable number of glioma tissues for studying the disease, the glioma bank was established. Methods The tumour tissues were collected during operation, including tumour, adjacent non-cancerous tissue and the serum of patients were obtained before operation. RNA, DNA and protein of the tissues were collected and stored with liquid nitrogen. A set of detailed information of the patients was recorded. Results Genomic DNA and RNA were extracted from the whole blood and tumor tissue of glioma patients, whose 0D260/280 ratios were between 1.8 -2.1. The protein was extracted from glioma tissues, which could meet the need of molecular biology research by the bicinehoninie acid (BCA) protein assay detection. Conclusion A certain size of glioma tissue bank and a set of related methods have been established and formed for supplying a service platform of glioma research in the future.
