安徽省首次分离培养布鲁氏菌方法的研究 被引量:5

First isolation and laboratory diagnosis of brucella in Anhui province

摘要 目的建立布鲁氏菌病的病原分离培养及鉴定方法,对安徽省一例疑似布鲁氏菌病病人进行实验室确诊。方法采病人急性期血液,血增菌液增菌后接种巧克力琼脂平板培养,培养菌株进行革兰氏染色、生化鉴定及16S rRNA序列测定分析。结果培养菌株革兰氏染色阴性、短杆球菌、无芽孢,VITEK2 Compact全自动生化鉴定仪报告为羊种布鲁氏菌(分辨率为99%),16S rRNA序列比对与布鲁氏菌相似度为100%。结论首次对病人血液分离到一株羊种布鲁氏菌,确定该病人患有布鲁氏菌病。 Objective To establish the method of isolation and identification of brucellosis pathogen, and confirm a case of suspected brucellosis patient in our province. Methods The acute blood of the patient was collected, then enriched in fluid and cultivated with chocolate agar plate. The cultivation of strains was Gram staining, biochemical identification and 16S rRNA sequences analysis. Results The strains were G- , short rod, and had no endospore. The report by VITEK2 Compact automatic biochemical identification instrument was brucella melitensis ( resolution was 99% ). The 16S rRNA sequences similarity was 100%. Conclusion A strain of brucella melitensis was isolated firstly from the blood sample of patient, and the patient suffered from brucellosis was determined in our laboratory.
