摘要
目的研究生长激素受体(growth hormone receptor,GHR)在结直肠癌组织及细胞系中的表达。方法应用免疫组织化学和Western blot检测16对结直肠癌患者的肿瘤组织及其配对癌旁对照生长激素受体的表达,用Western blot方法检测8种常见结直肠癌细胞系GHR的表达及其在细胞系中亚细胞定位。结果在结直肠癌组织中GHR的表达明显高于癌旁组织,具有统计学差异(P<0.05);在8种结直肠癌细胞系均检测到GHR的表达;GHR的分布主要位于结直肠癌细胞膜及胞核。结论生长激素信号轴在结直肠癌的发生、发展中可能起到一定的促进作用。
Objective To study the expression oI growth hormone receptor ((iHR) m the colorectal cancer tissues and cell lines. Methods The expression of GHR was assessed by immunohistochemistry and Western blot in 16 tissues with human colorectal cancer and 16 paired normal epithelium tissues. The expression level and subcellular localization of GHR in 8 common colorectal cancer cell lines was tested by Western blot and immunofluorescence. Results The expres- sion of GHR was significantly increased in the colorectal cancer tissue compared with that in the normal mueosa epithelium (P^0. 05). Universal expression of GHR was detected in 8 common colorectal cancer cell lines. Most GHRs were located in the cell membrane and cytoplasm. Conclusion Growth hormone signal may participate in the carcinogenesis and tumor development of colorectal cancer.
