摘要
目的:探讨苦参碱治疗前列腺癌的可行性及机制。方法:用不同药物浓度苦参碱诱导PC-3M细胞不同时间后,在相差倒置显微镜观察细胞形态;用MTT法测细胞的增殖抑制情况;用流式细胞仪(FCM)进行细胞凋亡率检测;RT-PCR法检测PC-3M细胞内bcl-2、bax基因表达水平。结果:用苦参碱干预后随着药物浓度增加PC-3M细胞株生长明显受抑制;在FCM上可见凋亡率逐渐增加;PC-3M细胞中bcl-2 mRNA表达水平逐渐下调,bax mRNA表达水平逐渐上调,且呈时间-浓度依赖性。结论:苦参碱体外能有效抑制前列腺癌细胞株生长,其机制可能与诱导细胞凋亡、下调bcl-2表达及上调bax水平有关。
Objective: To study the feasibility of treatment of prostate cancer using matrine and its mecha- nism. Methods: Add a certain concentration of matrine in human prostate cancer pc- 3m cell incubated together, using the method of MTT and flow cytometry to detect the changes in cell proliferation and apoptosis, using re- verse transcription polymerase chain reaction(RT- PCR) to detect the expression level of bcl- 2 and bax gene in human prostate cancer pc - 3m cell. Result: matrine can inhibit the growth of human prostate cancer pc - 3m cell obviously, and showing a time concentration - dependent; The rate of apoptosis increases in flow cytometry; bcl- 2 mRNA expression level gradually lowered and bax mRNA expression levels gradually raised. Conclusion:ma- trine can inhibit the growth of human prostate cancer pc - 3m cell in vitro tests, the mechanism may relate to in- duced apoptosis and down to bcl - 2 expression and up - regulation of bax level.
