益髓通经方抗EAN水溶性成分提取工艺优选

Optimization of Extraction Technology for Anti-EAN Water-soluble Constituents in Yisui Tongjing Prescription

摘要 目的:验证益髓通经方经醇提后的药渣抗实验性自身免疫性神经炎(EAN)的活性,并优化其提取工艺。方法:建立大鼠EAN模型,将造模成功的大鼠随机分成模型组、免疫球蛋白组和给药组,按相应方法给药,用电子显微镜观察各组大鼠周围神经病理形态学超微结构;以干膏质量和蛋白提取率的综合评分为指标,采用正交试验法,选取加水量、提取时间和提取次数为考察因素,优化益髓通经方中水溶性成分的提取工艺。结果:给药组大鼠髓鞘脱失的情况得到明显改善,有髓纤维髓鞘板层结构整齐,胶原纤维聚集及有髓神经纤维新生等改变;最佳提取工艺为加8倍量水提取3次,每次3 h。结论:益髓通经方水溶性成分具有治疗EAN的效果;优选的提取工艺操作简单、稳定性好,适合于产业化生产。 Objective:To verify anti-experimental autoimmune neuritis(EAN) activity of dregs in Yisui Tongjing prescription after being extracted by ethanol and optimize its extraction process.Method:EAN rats model was established,then these rats divided into model group,immunoglobulin group,administration group,these rats were administered by the corresponding method,pathomorphological ultrastructure of rats peripheral nerve was observed by electron microscope.With comprehensive score of dry extract quality and yield of protein as index,orthogonal test was adopted to optimize extraction process of water-soluble constituents in Yisui Tongjing prescription with the amount of water,extraction time and times as factors.Result:The situation of demyelination of the administration group was significantly improved,the structure of myelinated fibers myelin lamellar was neat,at the same time you could still see the change in aggregation of collagen fibers and the newborn myelinated nerve fibers.The best extraction process for water-soluble constituents in Yisui Tongjing prescription was:extracted 3 times with 8 times the amount of water for 3 h each time.Conclusion:Water-soluble constituents in Yisui Tongjing prescription had treatment effect for EAN;This optimized extraction technology was simple,stable and suitable for industrial production.
