体外筛选剪切DNA的DNA酶

摘要 为得到能够剪切单链DNA序列的人工DNA酶,通过体外筛选的方法,得到了一类DNA酶F-9.F-9的结构由两侧与底物结合区域和中间的酶活性中心组成;F-9在一定的Cu2+、Mn2+离子浓度下发挥催化作用,可以与底物结合形成二级结构.F-9可以剪切DNA序列,底物识别区域ACTG为保守序列.同时,对DNA酶的剪切位点作了进一步研究,证明F-9是一个多位点剪切的DNA酶. In order to obtain single-standed^-DNA-cleaving artificial DNA enzyme, we carried out in vitro selection and got the DNAzyme F-9 that consists of a catalytic domain flanked by two single-strand binging arms. F-9 DNA enzyme can cleave DNA in a certain concentration of copper and manganese metal ions. F-9 can cleave DNA sequence containing ACTG. Study of the splice site of DNA substrate found that F-9 undergoes cleavage at several closely-grouped sites. Fig 6, Tab 1, Ref 19
