摘要
目的:探讨健脾活血方对肝纤维化大鼠肝功能、血清肝纤维化指标、病理组织学及胶原表达的影响。方法:采用四氯化碳(CCl4)诱导大鼠肝纤维化模型,将Wistar大鼠随机分为5组:正常组(N)、模型对照组(M)、秋水仙碱组(Q)、健脾活血方高剂量组(H)、健脾活血低剂量组(L),均以灌胃的方式给药。治疗结束后,动物采血处死,分离血清进行肝功能和肝纤维化指标检测,取固定部位肝组织做HE染色和Masson染色,观察肝组织中胶原纤维表达情况。结果:健脾活血方治疗后,大鼠血清中ALT、AST、TBIL的含量明显降低,ALB的含量明显升高,HA、LN、PⅢP的含量明显降低,与模型组比较有显著性差异(P<0.01);HE染色和Masson染色显示健脾活血方可明显改善肝纤维化大鼠肝组织的病理损伤,显著降低肝组织胶原纤维的表达。结论:健脾活血方对CCl4诱导的大鼠肝纤维化形成具有明显的抑制作用,其机制可能与保护肝细胞和抑制胶原纤维合成有关。
Objective: To investigate the effect of Jianpi Huoxue Decoction on liver function,serum fibrosis indices,liver pathological changes and the expression of hepatic collagen fibers in rats with liver fibrosis.Methods: Hepatic fibrosis was induced by carbon tetrachloride hypodermic injection.The Wistar rats were randomly divided into 5 groups: normal group,model control group,colchicines group,high-dose and low-dose groups of Jianpi Huoxue Decoction.All medicine was given by intragastric administration.At the end of treatment,serum alanine amino transamine(ALT),aspartate transamine(AST),albumin(Alb) and total bilirubin(TBil),serum hyaluronic acid,laminin and precollagen Ⅲ were detected.HE and Masson staining of liver tissue were determined.Results: Jianpi Huoxue Decoction can obviously decrease serum levels of ALT,AST,TBIL,HA,LN and PCШ,improve the level of ALB of rats;HE and Masson staining showed that Jianpi Huoxue Decoction can obviously reduce the expression of collagen fibers of liver tissue in hepatic fibrosis rats.Conclusion: Jianpi Huoxue Decoction can effectively prevent the pathological process in hepatic fibrosis rats.The mechanisms might be related with the protective effects on hepatic cells and the inhibition on collagen fibers synthesis.
