摘要
目的:观察软坚消瘿颗粒对自身免疫性甲状腺炎(AIT)大鼠甲状腺组织凋亡蛋白的影响。方法:将猪甲状腺球蛋白与弗氏佐剂溶合免疫注射配合饮用高碘碘水建立AIT大鼠模型,采用电化学发光法检测大鼠血清TGAb、TpoAb水平,采用免疫组织化学法测定大鼠甲状腺组织凋亡蛋白Fas Fas-L Bax Bcl-2的表达。结果:TGAb、TpoAb模型组与正常组比较具有显著性差别(P<0.01)。Fas、Fas-L、Bax模型组与正常组比较具有显著性差别(P<0.01);各中药治疗组与模型组比较均具有显著性差别(P<0.01);软坚消瘿颗粒高剂量组与雷公藤组比较有显著性差别(P<0.01)。雷公藤组及各中药治疗组与正常组比较Fas、Fas-L、Bax表达依然略高,但与模型组比较,均明显降低。Bcl-2模型组与正常组比较具有显著性差别(P<0.01);各中药治疗组与模型组比较均具有显著性差别(P<0.01);软坚消瘿颗粒高剂量组与雷公藤组比较有显著差别(P<0.01)。雷公藤组及各中药治疗组与正常组相比较Bcl-2表达有一定差异,但与模型组相比较,均明显升高。结论:软坚消瘿颗粒通过改善凋亡蛋白的病理形态结构从而有效改善自身免疫性甲状腺炎的病理状态。
Objective:To observe the effects of Ruanjian Xiaoying granules on autoimmune thyroid inflammation rats through the change of apoptosis proteins.Method:Tg and IFA/CFA with drink iodine water were used to make animal mode.The level of autoantibodies was detected with the electrochemical luminescence method and the levels of Fas,Fas-L,Bax and Bcl-2 with immunohistochemistry methods.Results:TpoAb and TGAb were significantly higher in model group comparing with those in the normal group.Fas,Fas-L and Bax in the AIT group were significantly higher than those in the normal group;those in the different dosages of Ruanjian Xiaoying treatment groups were significantly lower than those in the AIT group;those in the higher dose of Ruanjian Xiaoying treatment group was significantly lower than those in the Tripterygium Wilfordii Hook treatment group.The Tripterygium Wilfordii Hook and the different dosage of Ruanjian Xiaoying treatment groups were less than those in the normal group but better than those in the AIT group.Bcl-2 in the AIT group was significantly lower than that in the normal group.Bcl-2 in the different dosage of Ruanjian Xiaoying treatment groups was significantly higher than that in the AIT group.The higher dose of Ruanjian Xiaoying treatment group had significance compared with the Tripterygium Wilfordii Hook treatment group.Bcl-2 in the Tripterygium Wilfordii Hook and the different dosage of Ruanjian Xiaoying treatment group was less than that in the normal group but better than that in the AIT group.Conclusion:Ruanjian Xiaoying granules can promote the autoimmune thyroid inflammation rats to be better through changing the level of apoptosis proteins.
