山茱萸颗粒对糖尿病肾病大鼠TGF-β_1/Smad3通路的影响 被引量:22

摘要 目的:观察山茱萸颗粒对糖尿病肾病大鼠TGF-β1/Smad3信号传导通路的影响,探讨其对糖尿病肾病的防治作用及其机制。方法:采用单侧肾切除加STZ注射法改良复制糖尿病肾病模型,实验分为正常组、模型组、治疗组(山茱萸颗粒高、中、低剂量组)、缬沙坦对照组,观察糖尿病肾病模型大鼠一般情况、生化指标、肾组织病理变化、免疫组化检测肾组织TGF-β1、Smad3蛋白的表达。结果:各治疗组一般情况、肾脏病理变化均优于模型组,同时TGF-β1、Smad3蛋白表达水平较模型组下调;山茱萸颗粒各治疗组空腹血糖(FBG)、血脂(TC、TG)优于模型组及缬沙坦对照组,其中高、中剂量组疗效无显著差别。结论:山茱萸颗粒可用于治疗早期糖尿病肾病,机制可能与其能抑制糖尿病肾病大鼠肾脏中TGF-β1/Smads信号传导通路的激活有关。 Objective:To explore the mechanism of Cornus Granules on protecting kidney by observing changes of signaling pathway of the TGF-β1/Smad3 of the DN rats.Methods:The experimental rats were divided into three groups:normal group,model group and treatment groups(high,middle and low dose of Cornus Granules and valsartan group).General state of health,some biochemical indicators,the pathological change of nephridial tissue and the levels of TGF-β1 and Smad3 were observed.Results:General state of health and the pathological change of nephridial tissue of the treatment groups were all better than those in model group,and compared with model group,the expressions of TGF-β1 and Smad3 in treatment groups were lower.The levels of FBG,TC and TG of all dose Cornus Granules groups were lower than those in model group and valsartan group,the high and middle dose groups had similar effect.Conclusion: Cornus Granules could be used to treat early DN,the mechanism may be related to restraining the activation of TGF-β1/Smads signaling pathway.
