妊娠期全氟辛烷磺酸盐暴露对胎鼠睾丸胚胎间质细胞雄激素合成的影响 被引量:1

Effects of perfluorooctane sulfonate exposure during pregnancy on testosterone synthesis in fetal Leydig cells of fetal rats

摘要 目的研究全氟辛烷磺酸盐(perfluorooctane sulfonate,PFOS)对睾丸胚胎间质细胞(fetal Leydig cell,FLC)合成雄激素的影响。方法将20只健康SPF级SD妊娠大鼠随机分成4组,分别为对照(Tween-20)组和低(5 mg/kg)、中(10mg/kg)、高(20 mg/kg)剂量PFOS染毒组,每组5只。于妊娠第12~19天采用灌胃方式进行染毒,染毒容量为2 ml/kg,每天1次,连续8 d。染毒结束后,观察雄性胎鼠睾丸FLC数量,检测睾丸内睾酮浓度、FLC中胆固醇脂蛋白结合受体(scavenger receptor class B member 1,SR-B1)、类固醇激素合成急性调节蛋白(steroidogenic acute regulatory protein,StAR)、胆固醇侧链裂解酶(cholesterol side chain cleavage enzyme,P450scc)mRNA的相对表达量。结果与对照组相比,中、高剂量PFOS染毒组雄性胎鼠睾丸FLC数量减少;高剂量PFOS染毒组雄性胎鼠睾丸内睾酮浓度降低(P<0.05);高剂量PFOS染毒组雄性胎鼠睾丸组织中StAR mRNA、P450scc mRNA的相对表达量下降(P<0.05),SR-B1 mRNA相对表达量显著下降(P<0.01)。结论妊娠期染毒高剂量PFOS可降低雄性胎鼠睾丸FLC合成睾酮的能力。 Objective To investigate the effects of perfluorooctane sulfonate (PFOS) exposure during pregnancy on testosterone synthesis in fetal Leydig cells (FLC) of male fetal rats. Methods Twenty SPF health pregnant rats were randomly divided into four groups: the negative control group and PFOS-treated groups at the dose of 5,10 and 20 mg/kg body weight respectively, five rats in each. The control group received an equal volume of Tween-20 as vehicle (2 ml/kg). PFOS were given from the 12th to 19th gestation day by gavage,once a day for eight consecutive days. The numbers of FLC was observed. Intratesticular testosterone concentration was detected by enzyme-linked immunosorbent assay (ELISA). The mRNA of steroidogenic acute regulatory protein (StAR) , cholesterol side chain cleavage enzyme(P450scc) and Scavenger receptor class B member 1 (SR-B1) were detected by real-time quantitative RT-PCR. Results Compared with the control group, the numbers of FLC decreased in mMe fetus of moderate and high dose PFOS groups. Intratesticular testosterone concentration in high dose PFOS group was significantly lower than that in the control group (P〈0.05). The expression of StAR mRNA and P450scc mRNA in high dose PFOS group were down regulated (P〈0.05). The expression of SR-B1 mRNA in high dose PFOS group was down regulated significantly (P〈0.01). Conclusion Gestational exposure by high dose PFOS can decrease the ability of testosterone synthesis in fetal Leydig cells of male fetal rats.
