摘要
以西府海棠成熟果实为外植体,消毒后取其种子进行培养获取无菌苗。以MS为基础培养基,应用双因素完全随机试验方法,研究了不同植物激素及其质量浓度配比对组培苗增殖、离体叶盘愈伤组织诱导及不定芽再生的影响,并研究了不同的IBA质量浓度及光照条件对生根的影响。结果表明:西府海棠最适增殖培养基为MS+6-BA 0.5 mg.L-1+IBA 0.2 mg.L-1,平均增殖倍数为6.30;最适离体叶盘再生培养基为MS+6-BA 4.0 mg.L-1+NAA 0.5 mg.L-1;最适生根培养基为1/2 MS+0.3 mg.L-1IBA;弱光比强光更利于组培苗的生根诱导。
Seeds from ripe fruits of Malus micromalus were used as explants to obtain aseptic seeding. The main purpose was conducted to study the influence of different phytohormone to shoot proliferation, callus induction and rooting of M. mi- cromalus. The optimum medium for shoot proliferation is MS+6-BA 0.5 mg·L^-1 +IBA 0.2 mg ·L^-1 With the average pro- liferation value of 6.30. The optimum medium for adventitious shoot regeneration is MS+6-BA 4.0 mg·L^-1 +NAA 0. 5 mg ·L^-1. The optimum medium for rooting is 1/2MS+0.3 mg ·L^-1 IBA. The roots induced under low light are better than those under high light situation.
