摘要
提出了高效液相色谱法测定复方维生素片中β-胡萝卜素含量的方法。样品皂化后经石油醚提取,提取液经旋转蒸干后用二氯甲烷溶解。以ODS-C18色谱柱(150mm×4.6mm,5μm)为固定相,甲醇-异丙醇(1+1)溶液为流动相,用紫外检测器于波长448nm处检测。β-胡萝卜素的质量浓度在0.10~50.0mg.L-1之间与峰面积呈线性关系,检出限(3S/N)为0.03mg.L-1。方法用于复方维生素片中β-胡萝卜素的测定,加标回收率在90.0%~95.0%之间,测定值的相对标准偏差(n=6)在0.86%~3.2%之间。
A method of HPLC for the determination of β-carotene in compound vitamin tablets was proposed. The sample was saponified and extracted with petroleum ether, and the extract was concentrated in a rotary evaporator and the residue was dissolved with CH2 C12. The β-carotene was separated on ODS-C18 column(150 mm× 4. 6 mm, 5μm) with mixed solution of methanol and isopropanol (1+1) as mobile phase, and determined at the wavelength of 448 nm with UV detector. Linear relationship between values of peak area and mass concentration of β-carotene was obtained in the range of 0. 10--50. 0 mg·L-1 , with detection limit (3S/N) of 0. 03 mg·L^-1. The proposed method was applied to the determination of β-carotene in compound vitamin tablets, giving values of recovery obtained by standard addition method in the range of 90.0%--95.0% with RSD's (n=6) in the range of 0.86%--3.2%.
