浒苔多糖对2型糖尿病大鼠脂代谢及脂联素mRNA表达影响 被引量:1

EFFECTS OF ENTEROMORPHA'S POLYSACCHARIDE ON LIPID METABOLISM AND ADIPONECTIN GENE EXPRESSION IN TYPE 2 DIABETIC RATS

摘要 目的研究浒苔多糖(Enteromorpha’s polysaccharide,EP)对2型糖尿病大鼠脂代谢影响。方法高糖高脂饲料喂养,结合注射小剂量链脲佐菌素(STZ)建立2型糖尿病大鼠模型,EP采用高(600mg/kg.bw)、中(300mg/kg.bw)、低(150mg/kg.bw)剂量进行干预。测定血脂水平、抗氧化指标;HE染色观察肝脏和胰腺组织结构;RT-PCR测定脂联素(adiponectin,APN)mRNA的表达水平。结果模型组精神萎靡,脱发、多饮、多食、多尿和体重减轻等T2DM症状明显,EP高、中、低剂量组2型糖尿病症状有所减轻。与空白组比较,模型组TC、TG、HDL-C和LDL-C明显较高(P<0.01)。与模型组比较,EP高、中、低剂量组TG明显较低(P<0.01),EP高、中剂量组TC、LDL-C明显较低(P<0.01),EP高、中剂量组HDL-C/TC比值较高(P<0.01)。EP高、中、低剂量组MDA明显较低(P<0.01),SOD较高(P<0.05),EP高、中剂量组GSH-Px较高(P<0.01,P<0.05),EP高、中剂量组脂肪APNmRNA表达较高(P<0.05),EP高、中、低剂量组肝脏细胞脂肪变性程度较轻,胰腺的病理损害较小。结论 EP对2型糖尿病大鼠的脂代谢具有一定的调节作用。[营养学报,2013,35(2):181-185] Objective To study the effect of Enteromorpha's polysaccharide (EP) on lipid metabolism in type 2 diabetes. Methods A model of type 2 diabetes in rats was established by feeding high sugar and fat diet and an injection of STZ. The rats were then given po daily 150, 300 or 600 mg/(kg bw) EP for 4 w. After 4w, serum TC, TG, HDL, LDL-C, MDA levels, SOD and GSH-Px activities were measured. The pathological changes of liver and pancreas were also observed by HE staining and the mRNA level of adiponectin (APN) in adipose tissue was detected by real time-PCR. Results The diabetic symptoms such as listlessness, hypotrichosis, polydypsia, polyphagia, polyuria, emaciation in EP groups were lighter than those in mode| group. Compared with the control group, the levels of TC, TG, HDL and in mode~ group were significantly increased (P〈0.01) . Compared with the model group, the levels of TG and MDA were significantly reduced (P〈0.01) in all EP groups: TC, LDL-C significantly reduced and HDL-C/TC increased in high and middle dose EP groups (P〈0.01). SOD activities were increased in high dose EP group (P〈 0.05). GSH-Px activities and mRNA level of APN were increased (P〈 0.05) in high and middle dose EP groups. The steatosis of hepatic cells and the pancreatic pathological damage were lighter in all EP groups. Conclusion EP can regulate lipid metabolism in type 2 diabetes.
