莪术不同品种含药血清对人胃癌细胞BGC823增殖、凋亡及核质比的影响 被引量:15

摘要 目的探讨莪术3个不同品种蓬莪术、广莪术和温莪术含药血清对人胃癌细胞BGC823增殖、凋亡及核质比的影响,为临床治疗胃癌及中药抗肿瘤复方中莪术药材的品种选择提供实验依据。方法体外培养人胃癌细胞BGC823,加入高、中、低剂量莪术不同品种含药血清,倒置相差显微镜下观察各组细胞形态。应用四甲基偶氮唑蓝(MTT)法检测胃癌细胞BGC823增殖抑制状况,流式细胞仪检测胃癌细胞BGC823凋亡,应用Image.pro.pius.5软件分析了各药物含药血清对胃癌细胞BGC823核质比的作用。结果与阴性对照组比较,温莪术大小剂量、广莪术大中剂量及蓬莪术中剂量含药血清在加药后48 h都能抑制胃癌BGC823细胞增殖(P<0.05),抑制率以温莪术大剂量最高(P<0.01)。温莪术对BGC823的凋亡抑制作用较高,与阴性对照组相比有显著性差异(P<0.05);温莪术大剂量含药血清组对胃癌BGC823核质比的作用较大,细胞出现核固缩,核质比较阴性对照组明显变小。结论蓬莪术、广莪术和温莪术含药血清均能明显抑制胃癌BGC823细胞增殖及凋亡,温莪术大剂量含药血清组对胃癌BGC823核质比的作用较大。莪术品种用于治疗胃癌时建议选用温莪术。 Objective To explore the influences of the medicated (Rhizoma Zedoariae) , including Peng' ezhu, Guang' ezhu and Wen serum of three cuhivars of Ezhu ' ezhu, on proliferation, apoptosis and nucleo-cytoplasmic ratio in human gastric carcinoma cells BGC823, and provide the experimental base for selecting cultivars of Ezhu in treatment of gastric carcinoma and anti-tumor compound formulas. Methods BGC823 were cultured in vitro and then the medicated serum of three cultivars of Ezhu was added in high dose, medium dose and low dose. The morphologic changes of BGC823 were observed with inverted phase contrast microscope. The effect of medicated serum on BGC823 was studied by using methyl thiazolyl tetrazolium test (MTF), flow cytometry and nucleo-cytoplasmic ratio analysis. Results Compared with negative control group, the proliferation of BGC823 was inhibited in high-dose and lowdose Wen' ezhu groups, high-dose and mid-dose Guang' ezhu groups and mid-dose Peng' ezhu group (P 〈 0.05 ) after 48 hours, and the inhibitory rate was the highest in high-dose Wen' ezhu group (P 〈 0.01 ). The apoptosis of BGC823 was significantly inhibited in all Wen'ezhu groups compared withnegative control group ( P 〈 0. 05 ). The high-dose Wen' ezhu group had higher effect on nucleocytoplasmic ratio of BGC823, and karyopyknosis and nucleo-cytoplasmic ratio were reduced significantly compared with negative control group. Conclusion The medicated serum of Peng' ezhu, Guang' ezhu and Wen'ezhu all can inhibit the proliferation and apoptosis of BGC823, and Wen' ezhu in higher dose has higher effect on nucleo-cytoplasmic ratio of BGC823. It is suggested that Wen'ezhu can be selected for treating gastric carcinoma.
