应用GC-MS技术测定细菌结构脂肪酸鉴定脂环酸芽孢杆菌的研究 被引量:1

摘要 脂环酸芽孢杆菌菌体细胞膜的主要脂肪酸成分是含6个或7个ω环状脂肪酸,将其转化为脂肪酸甲酯,通过皂化、甲基化后提取脂肪酸甲基酯,用气相色谱质谱仪测定脂肪酸的类型及含量,试验结果与笔者利用PCR方法和Ribroprinter核糖体基因分型结果相一致,可在实践中应用脂肪酸测定法鉴定脂环酸芽孢杆菌。 The main fatty acid of cell membrane of Alicyclobacillus is co-cyclic fatty acid, the number of which is 6 or 7. The co-cyclic fatty acid will be transformed into fatty acid methylester and then will be extracted fatty acid methylester after Saponification and Methylation. The ypes and content of fatty acids was determinated with Gas Chromatogra- phy Mass Spectrometer(GC-MS). The Experimental results of GC-MS are the same to the experimental results of PCR and automated ribotyping system (RiboPrinter). So identification of Alicyclobacillus by detecting fatty acids is more feasible in practice.
