摘要 目的 :探讨胰岛素样生长因子 -1受体 (IGF 1R)mRNA在低氧大鼠肺小动脉壁的变化。方法 :采用原位杂交方法检测低氧性肺动脉高压大鼠和正常大鼠肺小动脉壁IGF 1RmRNA的表达 ,用图像分析技术检测肺小动脉的形态改变和半定量技术检测IGF 1RmRNA的表达强度。结果 :低氧性肺动脉高压大鼠肺小动脉管壁增厚、管腔狭窄 ,反映管壁增厚的两个指标MT %和MA %均显著高于正常对照组 (P <0 .0 5 )。正常对照组大鼠肺小动脉壁IGF 1RmRNA有微弱表达 ,低氧组肺小动脉壁IGF 1RmRNA表达呈强阳性 ,低氧组表达强度显著大于正常对照组 (P <0 .0 0 1)。结论 :低氧情况下 ,肺小动脉壁IGF Objective The experiment was designed to elucidate the change in insulin like growth factor 1 receptor(IGF 1R)mRNA in the walls of pulmonary arterioles of hypoxic rat.Methods The expression of IGF 1R mRNA was detected by hybridization in situ technique on the walls of pulmonary arterioles in the rats with hypoxic pulmonary hypertension and normal. Morphological change in pulmonary arterioles was determined by image pattern analysis technique, the level of expression of IGF 1R mRNA was detected by semi quantitative technique.Results Compared with the normal control group, the walls of pulmonary arterioles were found to be thickened with both MT% and MA% higher significantly in the rat with hypoxic pulmonary hypertension ( P <0.05,respectively).The expression of IGF 1R mRNA of the pulmonary arterioles in the hypoxic rats was stronger than that in the normal( P <0.001).Conclusion In hypoxic condition, the level of the expression of IGF 1R mRNA in the pulmonary arterioles is closely related to hypoxic pulmonary vascular remodelling.
