摘要
目的 :阐明内皮素 (ET)与脊髓损伤 (SCI)后血脊屏障损害的关系 ,为临床治疗SCI提供指导。方法 :SD大鼠 2 4只 ,分为 4组 ,即生理盐水组、ET 1组、损伤 +生理盐水组和损伤 +PD1 4 5 0 6 5组。实验一 :无损伤组分别于鞘内注射生理盐水或ET 1。实验二 :压迫法致伤脊髓 (5 0 g ,1min) ,分别于伤前 1 0min鞘内注射生理盐水或非选择性ET受体拮抗剂PD1 4 5 0 6 5。伊文思兰 (EB)定量法评价血脊屏障功能 ,干湿重法测定脊髓中水含量。结果 :鞘内注射ET 1 1 0ng增加了脊髓组织中EB和水含量 (P <0 0 1 ) ,SCI后脊髓中EB和水含量明显升高 (P <0 0 1 ) ,PD1 4 5 0 6 5 (5 0 μg)降低了损伤脊髓中EB和水含量 (P <0 0 1 )。结论 :ET参与SCI后血脊屏障损害 。
Objective:To elucidate the relationship between endothelin (ET) and blood spinal cord barrier (BSCB) breach after spinal cord injury (SCI).Method: Sprayue Dawleys rats (n=24) were divided into four groups, i,e, uninjured with vehicle, uninjured with ET 1, Injured with vehicle and injured with PD145065. In the first study, uninjured groups were administered vehicle or ET 1 (10ng) intrathecally. In the second study, Injured groups received vehicle or PD145065 (50μg, nonselective ETR antagonist) 10minutes prior to SCI. The permeability of BSCB was evaluated quantitatively using Evans blue. Tissue edema was determined by percentage water content. Result: ET 1 increased the contents of Evans blue and water in the uninjured cord (P<0 01). The contents of Evans blue and water in the injured cord were higher than that of sham (P<0 01). PD145065 decreased the amount of Evans blue and water in the traumatized cord (P<0 01).Conclusion: ET plays a role in breakdown of BSCB following SCI.
