缺氧条件下DNA双链断裂修复机制变动及其意义

摘要 实体肿瘤中相当一部分细胞处于乏氧的微环境中。乏氧是细胞对放化疗抵抗的重要原因之一。DNA双链断裂是最严重的DNA损伤,也是放疗杀伤肿瘤细胞的分子基础。在缺氧的情况下,肿瘤细胞发生一系列适应性变化,双链断裂修复机制也随之受到影响。非同源末端连接修复(NHEJ)和同源重组修复(HRR)是最主要的两个DNA双链断裂修复途径。在缺氧情况下,NHEJ活性得到增强,而HRR活性则受到削弱。这一变化对肿瘤的生物学行为产生影响,同时也为我们提供了提高乏氧肿瘤细胞杀伤率的新的思考角度。
