摘要
目的比较两款体积排阻色谱柱——SRT与SRT-C系列SEC柱对样品分离的差异性,优化改进美洛西林钠聚合物测定的分析方法。方法采用高效液相色谱法(HPLC),选用SRT与SRT-C柱,以磷酸盐缓冲液为流动相,用紫外检测器(UV)进行检测,流速1.0mL/min、检测波长254nm、柱温25℃。结果应用SRT-C色谱柱,美洛西林钠聚合物可达到良好的检测效果。美洛西林钠浓度在0.625~20mg/mL浓度范围内,峰面积与保留时间存在良好的线性关系,R=0.999 9。并且在此浓度内样品的峰面积重现性较好。结论SRT-C型体积排阻色谱柱可对美洛西林钠聚合物进行更为快速的检测,检测效率比药典方法更高。
Objective The two size exclusion column separation between the samples were analyzed to improve and opti- mize the analytical method of the current Sino-US Luo Xilin sodium polymers determination. Methods High performance liquid chromatography (HPLC), SRT and SRT-C column with phosphate buffer as the mobile phase, UV detector (UV) were used to detec, flow rate was 1.0 mL/min, detection wavelength was 254 nm, column temperature was 25 ℃. Results The SRT-C column, mezlocillin sodium polymers could achieve good detection results. Mezlocillin sodium concentration was in 0. 625-20 mg/mL range, the polymer peak area and retention time had a good linear relationship, R=0. 9999. More over, in this con- centration within the sample peak area reproducibility was better. Conclusion SRT-C type size exclusion column mezlocillin so- dium may be more rapid detection of the polymer, and the detection efficiency is higher than the that of official method.
