摘要
目的:探讨熊去氧胆酸(UDCA)治疗妊娠期肝内胆汁淤积症(ICP)对胎盘多药耐药相关蛋白(MRP)的影响。方法:实验分为ICP未治疗组、ICP治疗组及正常妊娠组,测定三组孕妇分娩前及分娩后血清总胆汁酸(TBA)、甘胆酸(CG);逆转录实时荧光定量PCR和免疫组化法检测三组胎盘多药耐药相关蛋白(MRP1、MRP2、MRP5)的表达。结果:UDCA治疗显著降低TBA及CG水平(P<0.001);UDCA治疗显著下调ICP孕妇胎盘中MRP1 mRNA和蛋白的表达,显著上调胎盘MRP2、MRP5 mRNA和蛋白的表达(P<0.05)。结论:熊去氧胆酸可能通过调节胎盘多药耐药相关蛋白影响胆汁酸的转运来治疗妊娠期肝内胆汁淤积症。
Objective:To evaluate the effect of ursodeoxycholic acid(UDCA) on the multidrug resistance proteins(MRP) in the intrahepatic cholestasis of pregnancy.Methods: The experiment was divided into three groups,total bile acids(TBA) and glycocholic acid(CG) were detected before childbirth and after delivery.The expressions of MRP were determined by Real-time RT-PCR and immunohistochemistry.Results: UDCA treatment significantly reduced the levels of TBA and CG(P0.001).UDCA treatment significantly down-regulated the placental mRNA and protein expressions of MRP1,but up-regulated the placental mRNA and protein expressions of MRP2 and MRP5 in the pregnant women with ICP(P0.05).Conclusion: UDCA may adjust the placenta MRP to affect the transport of bile acid in the intrahepatic cholestasis of pregnancy.
