摘要
目的 通过多因素回归方式有效地探讨了抑癌基因 ,DNA以及AgNOR等分子指标对卵巢癌预后的影响。方法 应用PCR技术 ,免疫组化 ,图象分析和COX模型等先进手段进行分子基因水平的研究。我们对 10 0例卵巢组织标本包括 6 0例卵巢癌 ,2 0例卵巢良性肿瘤 ,2 0例正常卵巢组织进行联合检测。结果 通过COX多因素比例风险模型了解到抑癌基因P5 3AGNOR含量DNA倍体水平与卵巢癌预后密切相关。其中AGNOR含量为最相关因素。结论 COX模型为妇科肿瘤预后的筛选 ,提供了重要参数。
Objective The COX model was used to investigate the significance of abnormal expressions of p53,nuclear DNA content and AgNOR in the prognosis of ovarian cancers.Methods Using the techniques of PCR,immunohistochemistry and image analysis,we studied 20 cases of normal ovary tissues,20 cases of benign tumors and 60 cases of malignant epithelial tumors in the ovaries.The changes of p53 gene,DNA ploid level and AgNOR content,and the expression of p53 protein were measured.Statistically,the outcome of the experiment was evaluated with the COX model,and the three groups were compared with each other for the statistical purpose.Results For ovarian tumor the point mutation of p53 gene and the positive rate of p53 protein were significantly higher,but the level of aneuploid was lower.The number of AgNOR increased while its size became larger.All of these showed that there was a correlation between the above indexes and the 5 year survival rate of ovarian cancer.Conclusions The level of p53 gene expression,aneuplid DNA and AgNOR of ovary cancer tissues are correlated with the prognosis of ovarian cancer.The COX model may be used as a valuabel approach to analyze the prognosis of the cancer.
