摘要 目的 观察人类甲状腺癌中ret癌基因的活化及其意义。方法 应用RT PCR技术研究甲状腺癌新鲜组织中ret癌基因活化形式PTC基因的表达。结果  2 5例甲状腺乳头状癌中有 6例 (2 4% )PTC基因表达阳性 ,主要分布于Ⅱ级以上肿瘤中 ;而甲状腺滤泡型癌、甲状腺腺瘤和正常甲状腺组织中PTC均为阴性。结论 PTC基因是ret癌基因新的活化形式 ,ret基因的活化仅限于甲状腺乳头状癌类型中 ,可能是乳头状癌的特殊遗传事件 。 Objective: To investigate the frequency and significance of ret oncogene activation in human thyroid carcinomas. Methods: Expressions of PTC oncogene which is a novel rearanged version of the ret tyrosine kinase proto oncogene were detected in fresh tissues of human thyroid carcinomas by RT PCR. Results: We found the presence of an activated form of the ret oncogene in 6 (24%) of 25 papillary carcinomas. By contrast, none of follicular carcinomas, benige lesions and nomorl tissues of thyroid showed PTC positive. Conclusion:PTC oncogene is a novel activated form of ret oncogene and restricted to the papillary cancer subtype. From this findings, we would suggest that ret activation may reprsesnt a specific genetic event that may be of value in the differential diagnosis and determination of biologic behaviour of human papillary carcinomas.
