摘要
目的:研究胰腺癌中SIRT-1和Ku70的表达及其临床病理意义,为胰腺癌患者的预后及生物治疗提供理论依据。方法:采用免疫组织化学技术(S-P)法检测46例胰腺癌及20例非肿瘤性胰腺组织中SIRT-1和Ku70表达情况,并分析其与胰腺癌临床病理参数的关系。结果:胰腺癌组织中SIRT-1的表达明显升高,其表达强度与临床分期、淋巴结有无转移、年龄和肿瘤大小呈正相关(P<0.05),与组织学分级呈负相关(P<0.05),与患者的性别和肿瘤部位均无关。胰腺癌组织中Ku70表达明显升高,其表达强度与临床分期、淋巴结有无转移及呈正相关(P<0.05),与组织学分级呈负相关(P<0.05),与患者的性别、年龄、肿瘤部位和大小均无关。结论:SIRT-1和Ku70在胰腺癌组织中的表达显著高于非肿瘤胰腺组织,且与胰腺癌的临床病理及预后相关,在胰腺癌的发展中也起一定作用,存在协同关系。
Objective: To study the expression and clinical significance of SIRT-1 and Ku70 in pancreatic carcinoma, in order to provide valuable evidences for the prognosis and biotherapy of pancreatic carcinoma. Methods: The expression levels of SIRT-1 and Ku70 were detected by S-P immunohistochemistry in 46 cases of pancreatic carcinoma and 20 cases of non-tumorous pancreas tissue. Their relationship with clinic-pathological features and prognosis were also analyzed. Results: The expression of SIRT-1 and Ku70 were both significantly higher in pancreatic carcinoma higher than those in non-pabcreatic carcinoma pancreas. The expression of SIRT-1 was positively correlated with the age, size of tumor, clinical stages and metastasis of lymphnodes (P〈0.05) and negatively correlated with histologic grade (P〈0.05), while no significant difference of sex and the location of tumor was observed. The expression of Ku70 was positively correlated with the clinical stages and metastasis of lymph nodes (P〈0.05), and negatively correlated with the histologic grade (P〈0.05), while no significant difference of sex, age, the location and the size of tumor was observed. Conclusion: The positive expression ratios of SIRT-1 and Ku70 in pancereatic carcinoma were both higher than those in non-tumorous pancreas tissue, which were significantly related with the prognosis of patients. SIRT-1 and Ku70 may act synergistically in the development of pancereatic carcinoma.
