根癌农杆菌介导CMO基因在96-Ⅱ喜树中的遗传转化

摘要 利用根癌农杆菌介导法将抗旱基因——胆碱单加氧酶基因(CMO)转入到96-Ⅱ喜树体内,研究影响96-Ⅱ喜树遗传转化效率的因子,建立96-Ⅱ喜树遗传转化系统。结果表明:在遗传转化过程中,预培养3 d,将D600 nm为0.5的根癌农杆菌侵染叶片15 min,再共培养3 d,然后转入筛选培养基,有利于获得最高的遗传转化效率,经PCR和Southern blot检测,CMO基因已被整合到96-Ⅱ喜树的基因组中。
