摘要
目的观察0.1μmol/L白藜芦醇作用不同受体亚型(ERα+/ERβ+)MCF-7细胞的增殖情况,并探讨其作用机制。方法采用MTT法和流式细胞术检测白藜芦醇对细胞增殖及周期的影响;Western-blot法检测白藜芦醇与不同受体亚型结合后对细胞信号通路的影响。结果白藜芦醇作用后,细胞增殖活性ERα-ERβ+-MCF-7<MCF-7<ERα+ERβ--MCF-7;同时流式细胞术结果显示,ERα+细胞S期比例增加(P<0.05),而ERβ+细胞周期阻滞于G0/G1期。western-blot结果显示,白藜芦醇可提高ERα+细胞bcl-2、ERK-P和AKT-P蛋白表达,减少caspase-3、p38-P蛋白表达;对ERβ+细胞作用则相反。结论白藜芦醇对MCF-7细胞的双相调节作用可能与ER不同亚型亲和力有关。
Objective To observe the effect of resveratrol proliferation of different estrogen receptor subtypes of MCF -7 cells, and to study the mechanism. Methods MTT assay and flow cytometry were carried out to examine the abilities of cell prolifera- tion. Western blotting method was used to detect the changes of the signal pathway. Results The abilities of cell proliferation were ERα - ERβ + - MCF - 7 〈 MCF - 7 〈 ERα + ERβ - - MCF - 7. The S - phase of the ERα + cell cycle was increased by resveratrol, however, ERβ + ceils was arrested at phase G0/G1. The protein expression of bci - 2, ERK - P and AKT - P were increased in ERα +cells treated with RES. The effect of ERβ + cells was opposite. Conclusions The dual regulatory effect on MCF - 7 cells by resvera- trol may be related to the affinity of different estrogen receptor subtypes.
